Nope, if you donate or purchase Membership, access to the game by queue will be just the same as if you were a user.

Charmander is the biggest "challenge" early on, but you're also able to catch all** of the starter Pokemon later in the game.

I recommend You to start with water of course to beat the Brock easly,

then catch Pidgey with special stats and trade it for Odish - NPC Madeline, Viridian City.

Train it and You will have short battle with Misty too, 24-26 Gloom can solo second Gym Leader, watch out for Starmie.

Take some pots xD
